Kokkuvõte
Paljude PPM-tolerantsete endofüütiliste mikroorganismide levimus papaias, laia toimespektriga mikrobitsiid, mis on ette nähtud kasutamiseks taimsetes koekultuurides ja mis suudab varjatult ellu jääda MS-põhises söötmes, millel on mõju saastumise manustamisele. Taimede säilitusainete kombinatsiooni ™ kasutati papaia (Carica papaya) koekultuuride loomiseks piirkonna eksplantaatidest. Hinnates kolme tõeliselt kasulikku praktikat endogeensete mikroobide saasteainete kontrollimiseks, aksillaarsete võrsete soovitusi (1,0-1,5 cm) cv-st. Arka Prabhathi on töödeldud PPM ™ 5% abil neli tundi (T1), 50% 10 minutit (T2) või 100% 10 minutit (T3) ja kasvatatud MS-põhises papaiaasutuse keskkonnas (PEM). 4-6 nädala jooksul osutusid kõik ravimeetodid ebaotstarbekaks, sest kultuurid allusid nii mikroobsele saastumisele (80% T1 korral) kui ka fütotoksilisuse mõjule/saastumisele (90% T2 ja 95% T3 korral).
Ühe katsega vastuvõtmiseks mitmeetapiline korrus steriliseerimise abinõu (karbendasiimiks-tsetrimiidi-HgCl 2 ) vastu kultiveerides 0,05% PPM-täiendatud PEM kinnitas 35% nähtava bakteriaalse saastumise erinevalt 40% juhtimises. Ühendatud bakterikasvude üksikuid kolooniaid on uuritud 0,1% PPM-i sisaldava toitainegariga (NA), registreerides 60% isolaate kui PPM-i tundlikke. Kakskümmend PPM-i säilinud isolaati on valitud ja tunnustatud. See kinnitas 85% grampositiivset mikroorganismi koos 80% varjupaiga all olevate Firmicutes (55% eoseid moodustavate Bacillaceae ja 25% Staphylococcaceae) ja 5% aktinobakteritega ning 15% gramnegatiivsete proteobakteritega.
Umbes 50% isolaatidest jäi PEM-il kultiveerimisel täiesti ebaselgeks, samas kui ülejäänud kinnitatud aeglane edenemine ja paljud näitasid edusammude paranemist peremeeskoe ekstrakti lisamisel. Isolaatide kasvatamine PPM-ga täiendatud NA-ga näitas, et 90-95% talub 0,05-0,1% PPM ja 65% ületab 0,2% PPM. Sellegipoolest ei näidanud isolaadid PPM-ga täiendatud PEM-is ilmseid edusamme kohas, kus eosed moodustasid.
