Bestmanni-Ohira reaktiiv ja seonduvad diazoühendid asaheterotsüklite sünteesiks
Asaheterotsüklid on üks levinumaid ühendite rühmi, mida leidub arvukates bioaktiivsetes ühendites, looduslikes saadustes ja agrokeemiatoodetes, ja kahtlemata on uued juurdepääsumeetodid neile alati nõudlikud. Saadavate meetodite hulgas on diasoühenditega seotud 1,3-dipolaarsed tsüklilahendusreaktsioonid eriti atraktiivsed, kuna neil on võime regioselektiivselt kiiresti konstrueerida tihedalt funktsionaliseeritud asaheterotsükleid.
Selles kontekstis on Bestmanni-Ohira reaktiivist saanud tuntud reaktiiv 1,3-dipolaarsete tsüklilahendusreaktsioonide korral fosfonüülitud heterotsüklite tekitamiseks, lisaks selle laialdasele kasutamisele homologiseeriva ainena aldehüüdide muundamisel alküünideks.
Selles aruandes kirjeldatakse üksikasjalikult meie jõupingutusi Bestmanni-Ohira reaktiivi ja sellega seotud ühendite sünteetilise kasulikkuse laiendamiseks asaheterotsüklite, näiteks pürasoolide, spirooksindoolide, triasoolide, triasoliinide ja spiropürasoliinide valmistamiseks, rõhutades domino mitmekomponendilisi reaktsioone, kasutades kergesti kättesaadavaid lähteaine reaktiive.
Polümeeride atmosfäärirõhu plasma plasmajoaga töötlemine võimaldab biomolekulide reaktiivivaba kovalentset kinnitamist bioprintimiseks
• 3D-bioprintimine, kus rakke, hüdrogeele ja struktuurpolümeere saab kiht kihi kaupa keerukaks kujundada, lubab meditsiini ja biomeditsiiniteaduste edusamme. Põhimõtteliselt võimaldab see tehnika luua väga patsiendispetsiifilisi haigusmudeleid ja biomeditsiinilisi implantaate. Siiski puudub praegu võimalus trükitud komponentide, näiteks polümeeride ja hüdrogeelide, pinna biosobivuse ja liidese sideme kohandamiseks.
• Siinkohal demonstreerime, et atmosfäärirõhu plasmajoa (APPJ) suudab valkude ja hüdrogeeli reaktiivivaba kovalentse kinnitumise jaoks polümeersed pinnad lokaalselt aktiveerida üheastmelises protsessis soovitud kohtades. Polümeeridena kasutati polüetüleeni ja polü--kaprolaktooni (PCL). Valkude ja hüdrogeeli kovalentset seondumist demonstreeris resistentsus eemaldamise suhtes naatriumdodetsüülsulfaadi (SDS) vary pesemise teel.
• Immobiliseeritud valgu ja hüdrogeeli kihte analüüsiti Fourieri transformatsiooniga infrapuna (FTIR) ja röntgenfotoelektroonspektroskoopia (XPS) abil. Oluline on see, et APPJ pinna aktiveerimine muutis polümeeri pinnad ka kergelt hüdrofiilseteks, nagu on vajalik optimaalseks biosobivuseks.
Veekontaktnurgad olid stabiilsed vahemikus, kus biomolekulide konformatsioon on säilinud. APPJ-de ühe- ja kahekordse elektroodiga kujundusi võrreldi nende omaduste suhtes, mis olid olulised pinna lokaliseerimise funktsionaliseerimise, ploomi pikkuse ja kuju suhtes.
• Efektiivsuse tõendiks bioloogilises kontekstis kasutati rakkude adhesiooni ja proliferatsiooni paranemise tõendamiseks APPJ-ga töödeldud polüetüleeni, mis oli funktsionaliseeritud fibronektiiniga. Difluoroisoksasoolatsetofenoon: difluoroalküülimisreaktiiv 1,2-diketoonide orgaaniliste katalüütiliste vinüüloogsete nitroaldoolreaktsioonide jaoks
Difluoroisoksasoolatsetofenoon (DFIO) on välja töötatud uue difluoroalküülimisreagendina, mida saab hõlpsasti valmistada odavatest lähtematerjalidest. Kolesterooli mugavaks kolorimeetriliseks määramiseks, ilma et oleks vaja kromogeenset substraati, on välja töötatud paberipõhine testriba, mis koosneb tseeriumoksiidi nanoosakestest (nanotseeria) vesinikperoksiidist (H2O2) sõltuvatest värvi muutvatest nanosüümidest ja kolesterooli oksüdaasist (ChOx). Parandatud HPLC tingimused eumelaniini ja feomelaniini sisalduse määramiseks bioloogilistes proovides, kasutades ioonpaarreagenti
Eumelaniini ja feomelaniini leeliseline vesinikperoksiidi oksüdeerimine (AHPO), kaks peamist melaniinipigmentide klassi, annavad pürrool-2,3,5-trikarboksüülhappe (PTCA), pürrool-2,3-dikarboksüülhappe (PDCA) ja pürrool-2, 3,4,5-tetrakarboksüülhape (PTeCA) eumelaniinist ja tiasool-2,4,5-trikarboksüülhape (TTCA) ja tiasool-4,5-dikarboksüülhape (TDCA) feomelaniinist.
Nende viie markeri kvantifitseerimine HPLC abil annab kasulikku teavet melaniinide koguse ja struktuurilise mitmekesisuse kohta erinevates bioloogilistes proovides.
Nende markerite HPLC analüüsil, kasutades algupärast meetodit 0,1 M kaaliumfosfaatpuhvrit (pH 2,1): metanool = 99: 1 (PTeCA jaoks 85:15) pöördfaasilises kolonnis, oli mõningaid probleeme, sealhulgas kolonni lühike eluiga ja , välja arvatud peamine eumelaniini marker PTCA, kattusid teised markerid aeg-ajalt häirivate piikidega proovides, mis sisaldasid ainult nende markerite jälgi.

Sihtotstarbeliste fotoakustiliste pildistusvahendite loomiseks töötati välja lihtne fotoakustiline värvaine kui lihtne kasutada reaktiiv. Pliimolekul valmistati tõhusa kaheastmelise sünteesi teel odavast kaubanduslikult saadaval olevast lähtematerjalist. Värvaine kaasasündinud albumiiniga seonduvate omaduste tõttu on saadud tetrasiinist saadud värv võimeline kasvajas lokaliseeruma ja selle bioloogiline poolestusaeg on paar tundi, mis võimaldab optimeeritud jaotumisprofiili.
Tetrasiini olemasolu võimaldab omakorda siduda albumiini siduva optoakustilise signaalivahendi laia sihtmärgiks olevate molekulidega. Platvormi kasulikkuse ja kasutusmugavuse demonstreerimiseks loodi uus fotoakustiline proov kaltsiumi akretsiooni kuvamiseks, kasutades üheastmelist bioortogonaalset sidestusreaktsiooni, kus hiirtel saadi põlveliigese kõrge eraldusvõimega fotoakustilised kujutised juba üks tund pärast süstimine. Seejärel määrati kogu keha jaotus märgistades sondi 99mTc-ga ja tehes kudede loendamise pärast lahkamist.
