BrdU rakkude proliferatsiooni testi paketi tehing tuvastab rakulise DNA-sse sisseehitatud 5-bromo-2′-deoksüuridiini (BrdU) kogu rakkude proliferatsiooni kaudu, kasutades BrdU-vastast antikeha. Kui rakke kultiveeritakse märgistussöötmega, mis on kaasas BrdU-ga, on see pürimidiini analoog sisse ehitatud tümidiini asendajana prolifereeruvate rakkude äsja sünteesitud DNA-sse.
Pärast märgistuskeskkonna likvideerimist fikseeritakse rakud ja DNA denatureeritakse meie fikseeriva/denatureeriva vastusega. Seejärel lisatakse BrdU hiire mAb, et tuvastada sisseehitatud BrdU (DNA denatureerimine on hädavajalik, et tugevdada sisseehitatud BrdU juurdepääsu tuvastusantikehale). Seejärel kasutatakse kindla avastamisantikeha tuvastamiseks hiirevastast IgG, HRP-ga seotud antikeha. Värvi arendamiseks lisatakse HRP substraat TMB. Arendatud värvi neeldumise suurus on võrdeline rakkudesse sisseehitatud BrdU kogusega, mis näitab otseselt rakkude proliferatsiooni.

Taust
Halogeenitud nukleotiidid, nagu pürimidiini analoogi bromodeoksüuridiin (BrdU), on kasulikud tekkiva DNA märgistamiseks resideeruvates rakkudes ja kudedes. BrdU muutub tümidiini aseainena sisseehitatud replitseeruvaks DNA-ks ja sellele järgnev BrdU immunodetekteerimine, kasutades spetsiifilisi monoklonaalseid antikehi, võimaldab rakke tsükli S-s märgistada. Pärast rakkude või kudede impulsi märgistamist bromodeoksüuridiiniga võib BrdU (Bu20a) hiire mAb-d kasutada üheahelalise DNA sisseehitatud BrdU tuvastamiseks.

5-bromo-2-desoksüuridiin (BrdU) on pürimidiini analoog. Tümidiini asendajana saab see sisse vohavate rakkude äsja sünteesitud DNA-sse. Biovisioni BrdU rakkude proliferatsiooni testi paketipakett tuvastab sisseehitatud BrdU, kasutades hiire BrdU-vastast antikeha. Kindla BrdU-vastase BrdU-vastase antikeha tuvastamiseks kasutatakse hiirevastast HRP-ga seotud sekundaarset antikeha, mis võetakse vastu TMB (HRP substraat) lisamisega. Värvide muutmise ulatus on võrdeline rakkudesse sisseehitatud BrdU kogusega ja seda saab kasutada otse rakkude proliferatsiooni indikaatorina. Võrreldes täiesti erinevate rakkude proliferatsiooni testidega tuvastab see pakett ainult prolifereeruvaid rakke ja mitte mingil juhul külvatud rakke. See erakordselt õrn,

METABOLIC PROLIFERATION ANSAYS
Analüüsid, mis mõõdavad ainevahetust, on proliferatsiooni, elujõulisuse ja tsütotoksilisuse analüüsimiseks vastuvõetavad. Tetrasooliumisoolade, nagu MTT, XTT ja WST-1, odav hind värvilistele formazaaniühenditele või resasuriini bioredutseerimine toimub ainult metaboolselt energilistes rakkudes. Aktiivselt vohavad rakud parandavad oma metaboolset rongi, samas kui toksiinide eest katmata rakud võivad väheneda.
MTT rakkude proliferatsiooni testid
MTT (3- [4,5-dimetüültiasool-2-üül] -2,5-difenüültetrasooliumbromiid; tiasolüülsinine) on vees lahustuv tetrasooliumsool, mis annab kollaka vastuse, kui seda valmistatakse söötmes või soolavalikus, milles puudub fenoolroosa. Lahustunud MTT muudetakse lahustumatuks lillaks formazaniks tetrasooliumtsükli lõhustamisel dehüdrogenaasi ensüümide abil. Seda vees lahustumatut formazani võib lahustada, kasutades isopropanooli või täiesti erinevaid lahusteid, ja lahustunud varusid mõõdetakse spektrofotomeetriliselt, kasutades neeldumist kui ümberehitatud värvi fookuse funktsiooni.
XTT rakkude proliferatsiooni testid
Erinevalt MTT -st on XTT lõhustumisprodukt vees lahustuv; lahustamisetappi ei nõuta. Tetrasooliumsool XTT lõhustatakse keeruka rakumehhanismi abil formazaniks. See bioreduktsioon toimub ainult elujõulistes rakkudes ja seda selgitatakse NAD (P) H tootmiseks glükolüüsi teel. Formazan -värvaine tavaline kogus korreleerub tavapäraselt metaboolselt energiliste rakkude arvuga.
WST-1 rakkude proliferatsiooni testid
Tavaline tetrasooliumsool WST-1 lõhustatakse lahustuvaks formazaniks keeruka rakumehhanismi abil, mis esineb täielikult rakupõrandal. See bioreduktsioon tugineb peamiselt NAD (P) H glükolüütilisele tootmisele elujõulistes rakkudes. Formazan -värvaine tavaline kogus korreleerub tavapäraselt metaboolselt energiliste rakkude arvuga.
